The research in Taotao's lab lies at the interface of chemistry and biology, particularly, medicinal chemistry and chemical biology of metal complexes, including anti-cancer compounds with in vitro and in vivo activities, immunogenic cell death inducers, drug target identification, luminescent DNA probes, and drug self-assembly and delivery.

Moyi's paper entitled "Photoactivation of Boronic Acid-Prodrugs via a Phenyl Radical Mechanism: Iridium(III) Anticancer Complex as an Example" has been accepted by JACS. Well done!

Boronic acid (or ester) is a well-known temporary masking group for developing anticancer prodrugs responsive to tumoral ROS, but their clinic application is largely hampered by the low activation efficiency. Herein, we report a robust photoactivation approach that can spatiotemporally convert boronic acid-caged iridium(III) complex IrBA into bioactive IrNH2 under hypoxic tumor microenvironments. Mechanistic studies show that the phenyl boronic acid moiety in IrBA is in equilibrium with phenyl boronate anion that can be photo-oxidized to generate phenyl radical, a highly reactive species that is capable of rapidly capturing O2 at extremely low concentrations (down to 0.02%). As a result, while IrBA could hardly be activated by intrinsic ROS in cancer cells, upon light irradiation, the prodrug is efficiently converted into IrNH2 even in limited O2 supply, along with direct damage to mitochondrial DNA and potent antitumor activities in hypoxic 2D monolayer cells, 3D tumor spheroids, and mouse-bearing tumor xenografts. Of note, the photoactivation approach could be extended to intermolecular photocatalytic activation by external photosensitizers with red absorption and to activate prodrugs of clinic compound, thus offering a general approach for activation of anti-cancer organoboron prodrugs.

This paper summarizes our recent series works on stimuli-activatable gold prodrugs. It is hoped that this account will spur the creation of better approaches to advance gold complexes closer to clinical application.

The development of metal‐based anticancer drugs has been hampered, among other reasons, by their lack of selectivity for cancer cells. In a recent article, Zou and co‐workers presented the successful intracellular activation of organogold(I) complexes for potential cancer treatment through Pd(II)‐mediated transmetallation, overcoming some off‐target activity of novel gold‐based drugs. This unique strategy builds the perfect bridge between metallodrug usage and bioorthogonal intracellular catalysis for more advanced and selective therapies. Such an approach will hopefully pave the way for forthcoming studies in medicinal inorganic chemistry. (https://doi.org/10.1002/cbic.202100015)

  • Highlight in Angew Chem: A photoactivatable prodrug approach overcomes the deactivation of drugs based on gold complexes caused by off‐target binding to serum albumin. In their Research Article (DOI: 10.1002/anie.202000528), T. Zou et al. show that gold(III) hydride complexes are inert towards serum albumin in the dark. Once they enter tumor tissues, their thiol reactivity can be photochemically activated, thereby leading to potent inhibition of tumor thiol enzymes through the formation of Au−S bonds.

  • Light-controlled thiol-reactivity: Stable cyclometalated gold(III)-hydride complexes can photo-dissociate hydride ligand and form coordinative [Au-S] adducts, leading to potent suppression of thioredoxin reductase, up to >400-fold increase of cytotoxicity (vs dark) towards different cancer cells with little influence by serum albumin, and strong inhibition of angiogenesis in zebrafish models by one- or two-photon excitation.
